Properties: Calcium L-5-methyltetrahydrofolate (L-5-MTHF-Ca) is the calcium salt of L-5- methyltetrahydrofolic acid (L-5-MTHF), which is the predominant naturally occurring folate in foods. Folic acid (pteroyl glutamic acid, sometimes known as Vitamin B9), which is widely used for food fortification and as an ingredient in dietary supplements, is the precursor of L-5-MTHF. L-5-MTHF-Ca is intended for use in dry crystalline or microencapsulated form as an alternative to folic acid in foods, including dietary supplements. L-5-MTHF-Ca is a white to light yellowish, almost odourless, water-soluble crystalline powder. It is obtained by reduction of synthetic folic acid with sodium borohydride, condensation of the resulting tetrahydrofolic acid with formaldehyde, further reduction of the 5,10-MTHF with NaBH4 to L-5-MTHF, and finally crystallization as the calcium salt. Crystalline L-5-MTHF-Ca is stable on storage (48 months at 40 ºC and up to 75% RH) after micronizing, and when compounded into vitamin and mineral tablets. It appears to be better suited than folic acid for the vitamin enrichment of foods. L-5-MTHF-Ca in microencapsulated form, preferably with ascorbate as an antioxidant, has also been shown to have long-term stability in a variety of foodstuffs. Tests with L-5-MTHF-Ca have shown that L-5-MTHF has a similar or slightly higher bioavailability and bioefficacy than folic acid.

Folate is an essential nutrient that supports neural tube development during pregnancy. Over 30% of people have a genetic mutation to MTHFR that doesn’t allow their bodies to convert folic acid to folate. This means the only way to supplement folate is with L-5-MTHF as it is the exact form the body needs.
2.In Pharmaceutical Industry
1)L-Methylfolate (5-MTHF) in depression
L-methylfolate is becoming widely recognised as an effective complementary medicine for patients with severe depression. That is, using it alongside pharmaceutical drugs (SSRI) to enhance the overall effectiveness of treatment (much like fish oil and depression).
Several randomised clinical trials have shown that 15 mg/day of L-methylfolate calcium is a safe, effective and well-tolerated treatment for patients with major depressive disorder who experience partial or no response to SSRIs.
It appears to be particular effective in those with certain genetic variations (including MTHFR) and those with elevated inflammatory markers
2)L-Methyfolate (5-MTHF) in patients with diabetic neuropathy
Diabetic neuropathy is nerve disorder that develops over time in those with poorly managed type 1 or type 2 diabetes.
In a recent study, 544 patients with diabetic neuropathy were given Metanx (L-methylfolate-methylcobalamin-pyridoxal-5-phosphate) to trial for over a year. Based on self-reported surveys, overall pain rating decreased by 32% those previously treated with medications reported a 52% improvement in medication satisfaction.
Metanx is a combination vitamin B supplement, so this study did not test the effects of L-methylfolate on its own.
